Do all computers support booting from USB? How do I boot into USB? What happens if I go into bios and it doesn't show a USb option? Do i need to connect the USB first then select to boot from USB ?

No, not all computers support it. This can vary by motherboard; most new ones can.
Plug your flash drive in, reboot, and choose the flash drive from the boot menu.
You have to have something on the flash drive to boot from though...

what is a good way to test if my motherboard can boot up the flash drive?

What kind of things do i need to have in my flash drive that i can download to make it boot from there?

Just go into your BIOS and see if you can select USB as a boot device. If you can, then you can boot VIA USB, if you can't... well then you can't boot VIA USB.

It depends on what you're trying to boot...what are you wanting to boot into that you want to put on your flash drive?
