Default Lock/Protect a partition.

I have a 80gb hard drive I partitioned into two parts. One I am going to use to back up my system drive, and the other to transfer data around.

I would like to know if there is some way I can lock down the first partiton, protecting it from any other changes. Is there a program that will allow me to do this? Or is there some way to do it in windows? I would also like to use this as a usb boot device, so locking and unlocking should be able to be done easily and any where.

Is there any way to enter the password to grant access, in windows explorer, on my pc, and any pc this usb hard drive is plugged into?
This is in lock folder xp 3.5.
