Question HD partitions & how to use them

Ok I just build my 1st computer and use it primarily for gaming. I have a 250gb HD, everything is working fine. My question is about partitioning my hard drive. I think i understand why its benificial to do this, but I'm not sure just how to access the data on the different partitions. If I have say partition C, D, E, ect. with my OS on C and say my games are all installed on D and music and photos on E then do I need to somehow tell my computer where to look for the data or program? and if so then how do I do that?

and yes I plan on running Fdisk on my new HD before I reinstall XP and add partitions
