Can't Communicate Between Computer And Radio

I am trying to connect my computer to a radio so that I can computer control it with some software.
The connection is typically made from serial to serial, but since my computer doesn't have a serial port, I have a Prolific USB to Serial Cable cable connecting the two (a null modem cable is used also)
I am unable to get the computer to talk to the radio.
I am using Windows XP SP3.
In my Device Manager, under Ports, I have "Prolific USB-to-Serial Comm Port (COM4)" listed. (BTW ... COM4 is also identified as "Communications Port")
In my radio tuning software, I have chosen COM4 as the receiving comm port. (I was only able to select from COM4, COM6, COM7).
I have ensured that all the properties for communications in the radio software match those that the Com port possesses.

Without knowing anything about the software (I have asked for support from the software developer which believes it to be a hardware issue), is there something I should consider that may be preventing the communication?
