I would say that rule in general, when the heatsink is removed.

Yeah, you'll have to re-apply the thermal paste once you take the HSF off.

What can I use to take the stock thermal paste off the CPU and cooler?

Isopropyl alcohol + coffee cloth or a lintfree cloth.

Get them at your local drug store or whatever.

Yeah basically just rubbing alcohol, I've heard coffee filters are also helpful since they have no lint.
