Is it true that all spyware removers and anti-viruses can only scan and remove viruses and trojans that are present on the libraries and can't be able to detect those who are on the backdoors or are running with a legitimate application/programs?

I think he means (most) spywarekillers can't run in the background to scan for trojans and detect spyware running in memory.

I am not sure about other programs but webroots spysweeper can run in the background to detect spyware in memory

BTW, Sygate firewall detects if an application loads unknown .dll 's.
