will this

work with an ecs rs400a mobo and a 380 watt PSU? I have that extra 4 pin molex connector... does that mean my PSU is pci-express compliant?

That card requires no special connectors, and if you're running minimal devices (e.g. no multiple hard drives) and there's enough amps on the 12v rail, that card will run. But I would get a new power supply along with that card, just to be safe.

Yes, your mobo has a PCI-E x16 slot so it will work. Also, I would agree that you should probably be safe and get a 400+ watt PSU (better safe than sorry)
