Vertical Sync (VSYNC): portion of composite video signal that tells the receiver where to place the image in the top-to-bottom dimension.

Generally in the computer world, vsync means synchronizing the refresh rate with the frame rate. ie. if your resolution is 1024x768 @ 75hz, your frame rate would be limited to 75 fps.
 
It's definately safe to turn it off. Some people say games play smoother with VSync on, but I doubt you'd really notice a difference. If you're running any benchmarks, make sure you have it turned off.
